Process improvement in a commercial setting
We will explore how commercial manufacturing processes for vaccines / immunomodulators can be improved while maintaining product quality, regulatory compliance and supply reliability. The presentation outlines a risk-based lifecycle approach for identifying, implementing and measuring improvements.

  3. From 18:50 to 19:10

Ivo Tjalma - PhD candidate TU Delft

Cell-Tolerant Radial Affinity Chromatography for Capture of Monoclonal Antibodies from Unclarified Harvest
Cell-Tolerant Radial Affinity Chromatography (cTRAC) is a novel technology that uses packed beds of large-bead affinity resins that specifically bind a soluble product while allowing cells and debris to flow through, enabling direct capture from unclarified feed streams (Raedts, 2019). By integrating upstream and downstream processing, cTRAC has the potential to eliminate conventional clarification steps and thereby reduce product losses, product degradation, process time, footprint, and costs (Frej & Hjorth, 2018; Kinna et al., 2016).

This presentation will summarize results on the application of cTRAC to process unclarified mAb-containing CHO cell cultures, highlighting its feasibility as an integrated capture step and discussing key areas for further research and development.

  4. From 19:30 to 19:50

Louis Boon - CSO JJP Biologics

Nebaprubart for the treatment of IgA-mediated inflammatory disorders: Autoantigen-specific IgA; Innocent till proven guilty
Autoantigen-specific IgA autoantibodies closely correlate with symptoms severity in a subgroup of patients in multiple autoimmune and fibrotic diseases. The value of measuring these IgA autoantibodies is the observation that the presence of excessive IgA/autoantigen immune complexes results in continuous CD89-mediated activation of myeloid cells, leading to severe tissue damage. Activation of myeloid cells - especially of neutrophils - is a highly underappreciated and untargeted hallmark of autoimmune and fibrotic diseases. Clinical experience shows that interfering with the IgA/CD89 axis by nebaprubart, resolves IgA/autoantigen-induced inflammation and subsequent tissue damage in Linear IgA Dermatosis (LAD) and proves IgA to be guilty to the pathology.
